Thames Valley Police said the eastbound carriageway between junction 14 at Hungerford and junction 13 at Newbury, where the crash happened, would be blocked for "many hours".
The westbound carriageway was closed to allow three air ambulances to land but has since reopened.
Police are advising motorists to avoid the area if possible.
South Central Ambulance said
six ambulances and two units from the Hazardous Area Response Team were at the scene.
Karyn Romeis, who is stuck in the eastbound queue behind the crash, said motorists "have been out of their cars for over an hour" and are now getting out to walk on the opposite carriageway.

M4 closed due to worst accident I’ve ever seen in many years of driving. Avoid area and if you’re caught in the tail back be patient....I promise you don’t want to get past it until emergency services are finished.
